comment Fitting an imported .txt data
The problem is: You have two independent variables x,y and mathematica wants to insert the values of the data-matrix into those values to fit the parameters a,b,c,d,e,f, but your matrix is 647x647. So the question for me (and therefore also for mathematica) is: How to gather value-pairs to use for (x,y)? I would have expected a matrix with N rows and 2 columns, then it would be clear (like in the first example here: reference.wolfram.com/language/ref/NonlinearModelFit.html)

And why is your approach different then Binarize? For me, /. x_ /; x > # -> 0 looks like replacing values above x>.8 (or .99) with 0 might also be highly dependent on the illumination, isn't it?

Well, I came up with the very simple idea of just using Binarize instead of your proposed arithmetic, which also works :-) Dunno why I just didn't think of this simple approach, probably just missed the forest for the trees :D

Could be, yes, but unfortunately the simple way isn't always simple for a beginner into the new language - no experience how to best combine the different mathematica commands into a working code :(

Thanks gonna try it out tomorrow :-) But looks really promising: short, clean and effective!

@nikie: As stated above, I havent implemented it yet. I hope my further details will help to understand it better..

comment Implementation of Partial Least Squares (PLS)?
@george2079 I haven't tried anything because I thought there would exist an implementation somewhere and if you're completely new to mathematica, it's not that easy to implement it and I'm sure it'll be prone to errors...
